The Alpha TC1-9 cell line, a pancreatic alpha cell clone, was derived from the Alpha TC1 line, established in transgenic mice expressing the SV40 large T antigen oncogene under the rat preproglucagon promoter. Alpha TC1-9 exhibits MHC heterozygosity from both parental strains. Unlike the parent Alpha TC1, it specializes in producing glucagon and does not generate insulin or preproinsulin mRNA. Glucagon synthesis is individually inhibited by rat recombinant gamma-interferon or mouse recombinant interleukin-1 in Alpha TC1-9, without affecting DNA synthesis. These cells grow with poor attachment, often forming clusters that can become suspended. They have low viability and tend to clump, making accurate counting difficult.
