The H441 (NCI-H441) cell line, derived in 1982 from the pericardial fluid of a male patient afflicted with papillary adenocarcinoma of the lung, exhibits notable characteristics. Microscopically, these cells display an epithelial-like morphology with the presence of multilamellar bodies and cytoplasmic structures resembling clara cell granules. Cultured primarily in adherent monolayers, these cells also manifest rounded floating or loosely attached cells. Importantly, H441 cells possess a KRAS mutation (p.G12V c.35G>T) and demonstrate the ability to form colonies in soft agar with or without serum. Furthermore, these cells express both mRNA and protein for the major surfactant apoprotein (SP-A), rendering them pertinent for investigations in respiratory physiology.
