Description

HeLa9903 is a cell line derived from the well-known HeLa cervical tumor cells, specifically engineered for research into estrogen receptor (ER) activity. This cell line is stably transfected with two constructs: one expressing the human estrogen receptor alpha (ERα) and another comprising a firefly luciferase reporter that is regulated by an estrogen-responsive element (ERE) linked to a mouse metallothionein promoter. Notably, HeLa9903 maintains the epithelial morphology and adherent growth characteristics typical of HeLa cells without the use of viral vectors for gene introduction.

Why choose HeLa9903 from AcceGen?

Choosing HeLa9903 from AcceGen ensures access to a well-equipped lab staffed by professional operators who prioritize precision and expertise in cell line management. AcceGen implements strict quality control measures, guaranteeing that each cell line is mycoplasma-free and meets the highest standards for research reliability. Additionally, the cells undergo short tandem repeat (STR) identification, confirming their authenticity and genetic consistency.

Application

FOR RESEARCH USE ONLY

HeLa9903 serves as a powerful tool in biological research, particularly in assays that assess ER transactivation. It is instrumental in predicting the binding affinity of various chemicals to ERα, enabling studies on the agonistic effects of compounds like bisphenol A and the influence of phyto- and mycoestrogens on estrogenic responses. Additionally, this cell line facilitates investigations into the dimerization and transcriptional activity of ERα, which are crucial for understanding potential health risks associated with estrogenic compounds. Overall, HeLa9903 is vital for advancing research in endocrinology and toxicology.

Frequently Asked Questions

  • What is the relation between Hela9903 and Hela cell line?

The HeLa9903 is HeLa cell line that is stably expressing human estrogen receptor-alpha, reportor gene (estrogen response element linked to luciferase), and firefly luciferase gene (pGL3(Luc+)).

  • What plasmid are used for transfection?

    The introduced plasmids include

pGL3-TATA-EREx5-BSD: reportor gene (upstream sequences of Xenopus-derived vitronectin gene containing estrogen responce element was linked to firefly luciferase gene (Luc+))

pRC/RSV-hERalpha Kozak: human estrogen receptor alpha gene driven by RSV promotor.

  • What is the recommended culture medium of HeLa9903 cell line?

DMEM with 10% heat inactivated fetal bovine serum
