The MH-S cell line represents a continuous culture of murine alveolar macrophages (AM), initially derived from bronchoalveolar lavage cells of Balb/cJ mice via transformation with simian virus 40 (SV40). Characterized by their adhesion properties, lack of contact inhibition, and trypsin sensitivity, MH-S cells exhibit distinctive features. They express intracellular T-antigen and manifest DNA synthesis through the incorporation of 3H-thymidine, boasting a doubling time of approximately 48 hours, resulting in a doubling of cell numbers within 96 hours. MH-S cells maintain typical macrophage morphology, with over 98% esterase-positive cells, while being negative for peroxidase. They further express cell surface Ia and Mac-1 antigens and exhibit Fc receptor positivity, demonstrated by their capacity for rosette formation and phagocytosis of antibody-coated sheep erythrocytes.
