THLE-3 cells are a cell line derived from primary normal liver cells through infection with SV40 large T antigen. These epithelial cells were isolated from the left lobe of a donor and exhibit characteristics similar to normal adult liver epithelial cells. Notably, they are non-tumorigenic when injected into athymic nude mice and have near-diploid karyotypes while lacking alpha-fetoprotein expression. THLE-3 cells are capable of metabolizing benzo[a]pyrene, N-nitrosodimethylamine, and aflatoxin B1 into their ultimate carcinogenic metabolites that adduct DNA, indicating functional cytochrome P450 pathways. To maintain these cells, when confluent, they are subcultured using 0.05% trypsin or trypsin/EDTA at a ratio of 1:2 to 1:6, and they should be fed 2 to 3 times weekly.- ONLINE INQUIRY
